چکیده
Sulfonylureas, widely used as hypoglycemic agents in adults with type 2 diabetes, have neuroprotective effects preclinical models of central nervous system injury, and children neuropsychomotor impairments linked to neonatal diabetes secondary ATP-sensitive potassium channel mutations. In the human rodent retina, we show that glibenclamide-activated sulfonylurea receptor 1 (SUR1) is expressed retina enriched macula; also it colocalizes Kir6.2, cation transporter TRPM4. Glibenclamide (glyburide), administered at doses did not decrease glycemia, or injected directly into eye, protected structure function various retinal injury recapitulate pathogenic neurodegenerative events diabetic retina. The downregulation SUR1 using a siRNA suppressed glibenclamide on excitotoxic stress-induced cell death. include transcriptional regulation antioxidant genes. Ocular could be repurposed for retinopathy.

Polymeric Microparticles as Alternative Carriers for Antidiabetic Glibenclamide Drug
Background: Glibenclamide is a lipophilic drug widely used in type 2 diabetes treatment. However, its low bioavailability limits its use. Thus, novel formulations should be applied to improve the drug’s bioavailability. Objectives: This study aimed to develop alternative carriers for oral delivery of glibenclamide. For this purpose, two biocompatible polymers, poly(e-caprolactone) and poly(but...
متن کاملThe Novel Oral Drug Subetta Exerts an Antidiabetic Effect in the Diabetic Goto-Kakizaki Rat: Comparison with Rosiglitazone
The aim of the present study was to evaluate the potential antidiabetic effects of two-component drug Subetta and its components (release-active dilutions of antibodies to β -subunit insulin receptor (RAD of Abs to β -InsR) and to endothelial nitric oxide synthase (RAD of Abs to eNOS)) in Goto-Kakizaki (Paris colony) (GK/Par) diabetic rats. Subetta was administered orally for 28 days once daily...
متن کاملElectrochemical determination of the antidiabetic drug repaglinide.
The electrochemical oxidation of repaglinide has been carried out in Britton-Robinson buffer at carbon paste and glassy carbon electrodes. Repaglinide exhibits a well-defined irreversible oxidation peak over the entire pH range (2-11). Differential pulse voltammetry was used to determine repaglinide in pure form. The peak current varied linearly in the following ranges: 8.0 x 10(-7)-3.2 x 10(-6...
متن کاملEstrogens and neuroprotection in retinal diseases
Estrogens play a critical role in the normal growth and development of humans and in recent years our understanding of their effects in the central nervous system (CNS) have been advancing rapidly. It is now known that estrogens influence synaptic plasticity, brain development, and memory. In addition, estrogens have been shown to be neuroprotective in degenerative disorders. The understanding ...
متن کاملMicroemulsions as Antidiabetic Drug Delivery Systems
Glibenclamide is practically insoluble in water and its gastrointestinal absorption is limited by its dissolution rate. Therefore, to enhance the drug dissolution and its hypoglycemic effects, the drug was formulated in different microemulsion systems and in vitro/in vivo evaluated. Microemulsion systems were prepared by Water titration method in which surfactants and cosurfactants (S/CoS) were...
